Pitchfork function in Sonic hedgehog signaling
In vertebrates Sonic hedgehog (Shh) signaling depends on primary cilia and failure of cilia formation and mis-regulation of Shh signaling lead to embryonic defects and a variety of human disease. We previously identified Pitchfork (Pifo) as a protein essential for cilia disassembly (Kinzel et al., Dev Cell 19, 66-77, 2010). Pifo is expressed in regions of high Shh signaling in the mouse embryo, is activated by Shh agonist and suppressed by Shh antagonist in cell culture, suggesting that Pifo is a Shh target gene and might function in the Shh signaling pathway. In this regard, the aim of this project is the investigation of Pifo functions in the Shh pathway.
