Pitchfork function in mouse development
In vertebrates Sonic hedgehog (Shh) signaling depends on primary cilia and failure of cilia formation and missregulation of Shh signaling lead to embryonic defects and a variety of human disease. We previously identified Pitchfork (Pifo) as a protein essential for cilia disassembly (Kinzel et al., Dev Cell 19, 66-77, 2010). Pifo is expressed in regions of high Shh signaling in the mouse embryo, is activated by Shh agonist and suppressed by Shh antagonist in cell culture, suggesting that Pifo is a Shh target gene and might function in the Shh signaling pathway. The aim of this project is to investigate if Pifo is a direct Shh target gene and if Pifo functions in the Shh pathway.
