Animal Model Generation

The scope of the team is the implementation, development and further refinement of novel technologies for mouse model generation as well as for gene therapy approaches.

There is an increasing demand for more sophisticated animal models and genome editing approaches. To meet these demands, our team provides the expertise and infrastructure for a whole variety of genome editing approaches and offers support in concept and design of any transgenic model generation. For the embryonic stem (ES) cell based approach of generating transgenic mice, our ES cell culture lab provides ES cells, reagents and protocols for the generation of targeted mouse ES cells. The microinjection team is equipped and trained to perform blastocyst injections of ES cells and the subsequent embryo transfer.

This production unit is part of the EUCOMM and EUCOMMtools (The European Mouse Mutagenesis Consortium) programs as cornerstone of the International Knockout Mouse Consortium (IKMC) platform.
Besides the classical, ES cell based approach we are utilizing nuclease (e.g. TALENs and CRISPR/Cas9) microinjection into pronucleus of single cell embryos. For this, we have established a vector free system utilizing Cas9 ribonucleoproteine complexes as the standard procedure for mouse production. In addition, our portfolio includes tetraploid complementation, sperm freezing and in vitro fertilization for the IDG strain archive and the import of new strains into the IDG barrier mouse facility.
