Project Description:

Novel monoclonal antibodies for β-cell analysis and targeting

Regeneration of β-cells or replacement by β-like cells derived from stem cells holds great promise to prevent diabetes. In our lab, we generated a large set of novel monoclonal antibodies (mAbs) targeted against human pancreatic cell types. Around 140 mAbs were discovered which identify either pancreatic endocrine, exocrine, ductal or endothelial cells.

The mAbs generated have great potential as

1) tools to isolate pancreatic cells types and their different subtypes for in-depth characterization of pancreas organogenesis, homeostasis and disease

2) markers to characterize pancreatic cell heterogeneity

3) novel biomarkers of diabetes progression

4) a method to enrich for endocrine progenitors and mature lineages during human stem cell differentiation

5) as a tool to detect, quantify and image β-cells in vivo and

6) specific carriers for targeted drug delivery

These novel mAbs give us a unique opportunity to develop cell-type specific diagnostic and therapeutical tools.
